Effect Mechanism:


For Acetaminophen :
-Analgesic: Acetaminophen may act predominantly by inhibiting prostaglandin synthesis in the central nervous
system (CNS).
-Antipyretic: Acetaminophen probably produces antipyresis by acting centrally on the hypothalamic heart-regulating
center to produce peripheral vasodilation resulting in increased blood flow through the skin, sweating and heat loss.
-Caffeine is central nervous system stimulant


Pharmacokinetic :


Acetaminophen:
-Oral: Rapid and almost complete.
-Distribution:
In breast milk: Peak concentrations of 10 to 15 mcg per mL (66.2 to 99.3 micromoles /L) have been measured 1 to
2 hours following maternal ingestion of a single 650-mg dose. The half life in breast milk is 1.35 to 3.5 hours.
Half- life: 1 to 4 hours.
Time to peak concentration: 0.5 to 2 hours.
Elimination
Renal, as metabolites, primarily conjugates: 3% of a dose may be excreted unchanged.
Caffeine:
Caffeine is absorbed readily after oral administration Maximal plasma concentration are achieved in adults within
one hour and the plasma half-life is about 3 to 7 hours-caffeine is almost completely metabolized in the liver by
oxidation, demethylation and acetylation to various xanthine derivatives, which are excreted in the urine.

Store below 30 °C.

Indications :

 

Megafen Extra® is indicated for the temporary relief of pain and discomfort associated with headache, tension headache,
migraine headache, osteoarthritis, arthritis, cold & flu symptoms, toothache, dental procedures, muscular aches, sore
throat and period pain .Reduces fever.
 

Consumption during pregnancy and lactation:

 

Pregnancy Category C.
Acetaminophen and Caffeine is excreted in breast milk.
Checking with physician if you are pregnant or nursing mother.
 

Common side effects :

 


Acetaminophen
1- Those indicating need for medical attention:
Incidence rare:
Agranulocytosis; anemia ; dermatitis, allergic; hepatitis; renal failure; sterile pyuria; thrombocyto penia.
Caffeine:
Psychiatric disorders: insomnia and restless .
